The series opens with two young adults driving down the road past a store sign advertising Tru Blood. They stop at the store and after a short confrontation with a vampire they take off. Next, Sookie Stackhouse, a waitress at a local bar notices a vampire who walks in and sits down in a booth. After locking eyes with the vampire she confronts him about what he'd like to order. The vampire asks for Tru Blood but due to a lack of local vampire patrons there isn't any in stock. Later Sookie runs out of the bar after the vampire and two people that lured him outside. She finds the two draining the blood from the vampire. Vampire blood is known to enhance the healing process in humans and give a bit of a sex boost. Sookie saves the vampire, who introduces himself as Bill.

After dark and a couple are driving down the road. The girl looks over at the sleeping male and reaches a hand over to his lap. He startles awake and asks her what she is doing. She replies that she is bored.
He notices a sign indicating a store sells "Tru Blood" and wants her to pull over. She screeches to a halt and pulls into the parking lot.
The tv is showing a debate with Nan Flanagan, representative of the American Vampire League and Bill Maher. She is arguing for the civil rights of vampires. Nan mentions that the Japanese have created synthetic blood satisfies all vampires nutritional needs. After the creation of the synthetic blood vampires decided to make themselves known to mainstream society.
The couple laughingly approach the counter asking the clerk about their sale of Tru Blood. The guy states that he didn't think there were vampires in Louisiana. The clerk speaks in an ominous voice and tells the couple that New Orleans is a mecca for the vampire. The guy laughingly says, "Even after Katrina? Didn't they all drown?" The clerk tells the couple, "Vampires can not drown. Because we do not breathe." The couple look startled then the clerk laughs and plays his remark off as a joke.
The guy refers to the girl as Kelly and asks the clerk where they can score some "V Juice". Another patron dressed like a hunter in the store comes up behind the couple and says he didn't think the clerk's joke was funny. The guy tells the hunter he doesn't care what the hunter thinks. He turns back to the clerk and tells the clerk he can pay good money for some vampire blood. Kelly tells her boyfriend no. She states that she knew a girl in college who did vampire blood and clawed her own face off.
The hunter/patron tells the couple they need to leave. The guy turns around to get in the hunter's face and the hunter shifts to a vampire's face. The couple leave the store.
The hunter sets a four-pack of Tru Blood on the counter to buy. He informs the clerk that if he ever pretends to be one of them again he will kill him. The clerk nods his head.
A truck pulls into the parking lot of Merlotte's, a bar. Sounds of people's chatter overlap as a waitress serves drinks to the customers. She serves a beer to a guy who is heard to plead with the Lord about not drinking too much. She passes a couple's table and the woman is berating the man with her. She goes to a family and the teenage son is having a tirade about being stuck in the town and wanting to get out. The waitress tells the teen to get out soon because the longer he waits the harder it is. The teen starts rambling questions wanting to know how the waitress knew what he was thinking. He wants to know if he was thinking out loud, if the waitress could hear his thoughts. The waitress beats a hasty retreat from the table. It becomes clear that none of the "heard" conversations were actually occurring out loud.
The waitress pauses, inhaling deep breaths and all conversation ceases. The only sounds in the bar are murmuring voices and the music in the bar. The waitress smiles and begins walking again through the bar.
A heavyset woman enters a store wearing an outlandish, garish outfit. She approaches a young black woman and inquires about sheets of plastic. The clerk tells her they don't sell the plastic. She suggests Home Depot. The woman stammers, saying she doesn't know what it's called but she drove over an hour for the plastic stuff. The clerk says they don't carry the stuff that woman doesn't even know the name of. The woman informs the clerk that the store's website says it is the most well-stocked store in five parishes and she drove over an hour to get there.
The clerk stands and asks the woman if their website had a phone number. She tirades at the woman wanting to know why the woman didn't call first or order the stuff online and have it delivered to her home. The woman demands to see the manager and calls the clerk rude. The clerk yells for the manager, tells the woman she is not rude, she is uppity. The manager comes over referring to the woman as Tara. Tara throws her employee vest at the man and tells him she quits.
The phone rings at Merlotte's and the bartender, Sam, hands the phone to the waitress. He calls the waitress and tells her Tara is on the phone. Tara tells her she quit her job. Tara tells Sookie she is coming over, she needs a Margarita. A big one. Sookie chides Tara for using the "J" word when Tara says "Jesus".
Mack and Denise Rattray sit down in Sookie's section. Mack is thinking inappropriate thoughts about Sookie and Denise is thinking sarcastic, nasty thoughts about her. Sookie turns in the Rattray's order to the cook, Lafayette Reynolds. She tells him if he drops some of the onion rings on the floor it will be ok with her. She takes Lafayette to task for what she calls "nasty talk".
A couple are having sex in the living room when the man stops. He points to two marks on her leg and asks if she has had sex with a vampire. She tells him once. She tells the guy she was broke and the vampire gave her a lot of money. The man sits up and asks her if she is a hooker. He tells her he doesn't pay for it. She tells him no she isn't but the vampire gave her a $1,000. He asks her what it was like. She tells him she likes it rough but the vampire was way too rough. He begins retreating from her. He searches for his clothes and she tells him she videotaped herself with the vampire. She asks if he wants to watch it. He throws his pants aside.
Tara is in the bar, drinking her Margarita and complaining about her life. She asks Sookie why she can't keep a job. Sookie tells her it's because she can't keep her mouth shut. Tara gets indignant and replies, "Who asked you?"
The bartender, Sam, asks Sookie how she is doing. He asks if there is anything he can do to improve the night for her. Sookie acts flustered and awkward. She looks around the bar and notices a guy sitting by himself. She stares at him. He stares back at her and she turns to Sam excitedly declaring she thinks Merlotte's has it's first vampire. She tells Sam she has been waiting for this moment since vampires came out of the coffin two years earlier. She goes over to the guy to take his order.
The man asks for synthetic bottled blood. Sookie tells the man they don't have any, that he is their first vampire. She asks if he drinks anything else. He tells her no but she can bring him a glass of red wine so it looks like he has a reason to be in there. Sookie leaves to get his order.
Mack and Denise introduce themselves to the vampire. They join him in his booth. Sookie and Tara are gossiping about the vampire sitting with the Rattrays. Sookie is upset, wanting to know what the couple are up to. She approaches the booth and hears the Rattray's thoughts. Denise is sizing up the vampire and thinking about draining and selling the blood of the vampire.
Sookie goes back to ask Tara and Sam for help. Tara tells Sookie no and Sam tells Sookie that vampires can take care of themselves. Sookie turns back to the booth and sees the Rattray and the vampire are gone. She runs outside after them. Sam takes off his apron and asks Tara if she knows how to tend bar. She tells him no. He throws her the apron and tells her to fake it. He runs after Sookie.
Sookie grabs a chain from the back of a pickup in the lot and follows the sounds of the Rattray's thoughts. They have the vampire incapacitated on the ground and are draining his blood.
Sookie sneaks up behind Mack and hits him with the chain. A woman is watching everything from the bushes. Mack turns and pulls out his knife. He lunges at Sookie with it and she throws the chain in his direction. The chain wraps around his throat. The woman watching in the bushes looks intently at Sookie and Mack, eyes glowing. The chain tightens and he drops the knight. The woman in the bushes runs away.
Denise runs toward Sookie and Sookie grabs Mack's knife. Denise tells Sookie she doesn't want to get on her bad side. Sookie replies that she doesn't think Denise has any other side. Denise turns back to the vampire and Sookie tells her she can't have the blood. Denise grabs Mack and tells Sookie things aren't over between them. The couple leave.
Sookie tries to tend to the vampire when Denise barrels toward her behind the wheel of a truck. Sookie pulls the vampire to safety. She removes the silver chains from his body that were holding him down. A dog rushes up to Sookie and the vampire barking. The dog licks Sookie's cheek, barks at the vampire and takes off.
Sookie tries to help the vampire and he jerks his arm. She stands up telling him she doesn't think he likes being saved by a woman. She looks down at the vampire and opens her thoughts trying to read his. Surprised she looks down and tells him she can't hear him. She grabs his face and looks into his eyes. She is pleased to realize she can't hear his thoughts.
The vampire asks her if she is afraid to be alone with a hungry vampire. She tells him no. He tells her that vampires often turn on those who trust them. She backs up and replies that humans turn on those who trust them as well. She tells the vampire she isn't a total fool and wraps the silver chain around her neck. He tells her she has other juicy arteries and mentions one in the groin area. She tells him that he might be a vampire but when he talks to her he will speak to her like the lady she is.
The vampire asks her if she wants to drink the blood that was collected. Sookie acts shocked and tells him no. The vampire tells Sookie he understands vampire blood makes humans feel more healthy and improves their sex life. Sookie tells him that she is healthy as a horse and has no sex life to speak of. She closes her mouth embarrassed. He tells her she can sell it and she tells him she wouldn't touch it.
The vampire asks her what she is. Sookie tells him she is Sookie Stackhouse, a waitress. She asks his name. He tells her it is Bill. She laughs at him such a common name. She asks him about the silver hurting him. Bill asks her to keep the information to herself. He tells her that vampires don't like publicizing their weaknesses. Sookie nods her head and tells him she needs to get back to work.
Sookie is nearing the door of the bar and Sam is waiting outside for her. He is panting and out of breath. He asks how she is. She tells Sam she is fine and that not all vampires can take care of themselves.
Sam thanks Tara for helping out in the bar. Tara wants to know how much Sam is going to pay her. He hands a twenty dollar bill. She asks him how he expects her to work for twenty dollars a night. Sam tells her she was just filling in. Tara asks about a job and Sam cuts her off. He tells her it would only be a matter of time before she went off on someone. Tara tells Sam she only goes off on stupid people. Sam tells her most of his customers are stupid people.
Tara tries a different method and tells Sam she can help him keep an eye on Sookie. She mentions how Sookie was looking at the vampire. Sam hires her.
The man who was having sex with the woman who has vampire bites comes into the bar. He asks for his sister. His name is Jason Stackhouse, Sookie's brother. Sam tells Jason that Sookie left before Jason got there.
Sookie lives with her grandmother and tells her a vampire came into the bar. The next morning Sookie's grandma tells Jason and Sookie a woman was found murdered, strangled. The woman's name was Maudette Pickens. Jason mentions that Maudette was a fangbanger. Sookie wants to know how Jason knows Maudette slept with vampires. He tells her he just knows things.
Sookie approaches Jason trying to read his thoughts. He cuts her off telling her he is her brother and she doesn't do that to him. Jason goes back to work.
The police come to Jason's job site. They ask Jason about his relationship with Maudette Pickens. He tells them he had no relationship with Maudette. They ask if he had ever been to her apartment. He tells them no. He says he can do better than Maudette Pickens. They ask if he was at her apartment the night before. He tells them yes he was. He said he hooked up with Maudette and they had sex. They ask how the sex was. They tell Jason they saw the videotape of Jason and Maudette having sex. They tell Jason he needs to come with them.
Sookie's grandmother asks how old she thinks the vampire is. Sookie tells her she doesn't know. Grandma asks Sookie to find out if he was alive during the Civil War. Sookie tells her the vampire would have a hard time going to the library at Noon to discuss the war if he had been alive during that time. Grandma tells her they would have a special meeting and Sookie says she will ask him.
Tara is ranting in the bar at people. She tells a customer not to snap at her, she has a name. Lafayette asks Tara what Sookie thinks of Jason being arrested for Maudette's murder. Tara tells Lafayette that Jason can do much better than Maudette. Lafayette calls her out for her crush on Jason.
Sookie arrives at work to find out that Jason had been arrested. Rene Lenier and Hoyt Fortenberry, Jason's coworkers, tell Sookie about Jason's arrest. Sookie turns around to find Vampire Bill in the bar. She approaches him blocking out the thoughts of everyone in the bar. Bill asks her what she is. She reminds him she already told him she is a waitress. He tells her she is something more than human. Bill asks her if he can call on her sometime. She asks him to meet her at the bar when she gets off work.
Bill doesn't want to cause her trouble at work so he leaves. Sam pulls Sookie into his office and begins to read her the riot act for associating with the vampire. Tara busts into Sam's office and joins in the conversation. Sookie has had enough of both of them and reminds them she can take care of herself. She leaves the office.
The bar closes and Sookie waits in the lot for Bill. Sam locks up the bar and asks Sookie if she wants him to wait with her. She declines his offer. She notices a car in the lot and realizes it is Mack and Denise Rattray. They jump her from the bushes and begin beating Sookie up. Denise kicks her in the face and she spits blood.
